The context

Jesus has just driven traders from the temple courts. Officials demand a sign. He answers with an imperative about destroying the temple and a promise to raise it in three days. The narrator adds, in verse 21, that he was speaking of the temple of his body, and that the disciples understood only after the resurrection.

What it establishes

It establishes an explicit identification between Jesus' body and the temple, and a prediction of his raising. The saying is offered as the sign his questioners asked for, which places the resurrection at the center of his answer.

What it does not establish

It does not establish that Jesus threatened the building. The imperative is addressed to his opponents and is conditional in force: they will destroy, he will raise. Nor was the saying understood at the time. The Gospel is explicit that its meaning came later.
